Susceptibility to Blocking by Lyt Antisera Clones: Specificity, Cell Surface Phenotype, and Characterization of Interleukin 2-dependent Cytotoxic T-Cell

Cytotox ic T-cel ls were derived from the per i toneal cavity of a C 5 7 B L / 6 mouse immunized with B A L B / c sarcoma Meth A and from the sp leens of B A L B / c x C 5 7 B L / 6 F1 (hereaf ter cal led CB6F1) mice immunized with B A L B / c leukemia R L ~ I . The cel ls were cu l tured in inter leukin 2 and c loned by limiting di lut ion, and their speci f ic i ty was determined by d i rect tests and compet i t ive inhibi t ion assays. C 5 7 B L / 6 ant i -Meth A ef fector cel ls recogn ized H-2D ~ determinants . CB6F1 anti-RLc;1 ef fector cel ls recogn ized a un ique cell sur face ant igen of leukemia RLS'I. The speci f ic i fy was mainta ined in long-term cul ture. The cell sur face pheno type of the c loned ef fector cell l ines as determined by absorp t ion analys is was Thy-1 .2 § Lyt1.2 § 2.2 § and 3.2 +. Cytotox ic i ty was b locked at the target cell level by ant isera against H-2D ~, but not H -2D" or H-2 b, and at the ef fector cell level by ant isera against Lyt -2.2 and 3.2, but not Lyt-1.2, Ly-5.1 or Thy-1.2 .
